It doesn't take a very long time to get up to the tomb, and it probably only takes 30 - 40 minutes to go around the tomb. I would say that you should allow 1 - 1.5 hours for the whole trip up there. It can be sorted quite simply with one of the taxis in the main car park at the entrance to the Valley of the Kings. You need to get your ticket for Ay from down there as well. I hope that helps.
